Description

Alprazolam is a short-acting triazole benzodiazepine that produces its inhibitory actions in the central nervous system, especially at the level of the limbic system. It is assumed that much or perhaps all of the effects of benzodiazepines are the result of inhibition mediated through the neurotransmitter amino acid, gamma-aminobutyric acid (GABA). This action is a consequence of its binding to specific receptors (benzodiazepines) that are closely related to GABA receptors and transmembrane chloride ion channels. Its anxiolytic effect is the most notable, although it also has hypnogenic, muscle relaxant and anticonvulsant properties.
It is well absorbed from the GI tract, reaching peak plasma concentrations in 1 to 2 hours; these concentrations are directly proportional to the doses administered. It is highly bound (80%) to plasma proteins and is distributed appropriately in the body. It crosses the blood-brain barrier and the feto-placental barrier. It is biotransformed in the liver, where alpha hydroxy-alprazolam and a benzophenone are formed; the biological activity of the former is almost half that of the original drug, and the latter is inactive. Alprazolam and its metabolites are excreted in urine and breast milk. Its half-life is 12 to 15 hours.
